چکیده

Curcumin is known could inhibit the activation of NF-kB, which is important in osteoclast differentiation and activation. The aims of this research were to conduct the effect of curcumin and Pentagamavunon-0 or PGV-0 [2,5-bis-(4’-hydroxy-3’methoxybenzylidene)-cyclopentanone], on osteoclast differentiation and apoptosis. Osteoclast differentiation was carried out using Osteoclast Differentiation Factor (ODF) induction model in Raw 264.7. The evident of osteoclast was determined microscopically using inverted microscope after TRAP staining. The apoptosis of osteoclasts were determined microscopically using confocal microscope. The results showed that PGV-0 and curcumin at the concentration of 5 μM and 10 μM, respectively inhibited osteoclast formation by 50 –80%. PGV-0 and Curcumin induced apoptosis on osteoclast cells at the concentration of 10 μM and 20 μM respectively. Those effects were dose dependent.

Immunological Reaction in TNF-α-Mediated Osteoclast Formation and Bone Resorption In Vitro and In Vivo

Tumor necrosis factor- α (TNF- α ) is a cytokine produced by monocytes, macrophages, and T cells and is induced by pathogens, endotoxins, or related substances. TNF- α may play a key role in bone metabolism and is important in inflammatory bone diseases such as rheumatoid arthritis. Cells directly involved in osteoclastogenesis include macrophages, which are osteoclast precursor cells, osteobla...

Effect of Curcumin on Doxorubicin-induced Cytotoxicity in H9c2 Cardiomyoblast Cells

Objective(s) Doxorubicin (DOX), a widely used chemotherapeutic agent can give rise to serve cardiotoxicity by inducing apoptosis. Curcumin, the active compound of the rhizome of Curcuma longa L. has anti-inflammatory, antioxidant and anti-proliferative activities. Curcumin has been identified to increase cytotoxicity in several cancer cell lines in combination with DOX, but there is no study a...

Synthesis Pyrano [2,3-c] Pyrazole-based compounds to induce apoptosis by reducing the expression of anti-apoptotic Bcl-2 protein in human breast cancer MCF-7 cells

Aim: Bcl-2 is a potential target for tumor treatment. The inhibition of the Bcl-2 production is research target of attract in the field of anti-cancer drug development. Recently, the assessment of antitumor activity appeared to be promising for pyrazole derivatives. Therefore, this study was designed to investigate the anti-cancer effects of novel pyrazole derivatives (HN1and HN2.). Material an...

The Modulatory Effects of Mesenchymal Stem Cells on Osteoclastogenesis

The effect of mesenchymal stem cells (MSCs) on bone formation has been extensively demonstrated through several in vitro and in vivo studies. However, few studies addressed the effect of MSCs on osteoclastogenesis and bone resorption. Under physiological conditions, MSCs support osteoclastogenesis through producing the main osteoclastogenic cytokines, RANKL and M-CSF. However, during inflammati...

LIS1 Regulates Osteoclastogenesis through Modulation of M-SCF and RANKL Signaling Pathways and CDC42

We have previously reported that depletion of LIS1, a key regulator of microtubules and cytoplasmic dynein motor complex, in osteoclast precursor cells by shRNAs attenuates osteoclastogenesis in vitro. However, the underlying mechanisms remain unclear. In this study, we show that conditional deletion of LIS1 in osteoclast progenitors in mice led to increased bone mass and decreased osteoclast n...
